Researchers develop novel methods for treating Clostridium difficile infections

Researchers at Brigham and Women′s Hospital (BWH) and Massachusetts General Hospital (MGH) have identified the metabolic pathways that Clostridioides difficile employs to enter the gut quickly.

 

New prevention and treatment strategies for the most common hospital-acquired infections (HAIs) types and causes of antibiotic-associated diarrhea are suggested by the findings. The results are published in the journal Nature Chemical Biology.

 

Researchers examined the real-time metabolism of living cells under anaerobic conditions using a technique known as high-resolution magic angle spinning nuclear magnetic resonance spectroscopy (HRMAS NMR). The team created a method to measure carbon and nitrogen transport across anaerobe metabolism simultaneously and used computational simulations to detect metabolic changes in C. difficile as nutrition availability declined. The researchers found that C. difficile initiates the fermentation of amino acids to jump-start its metabolism before activating pathways to ferment simple carbohydrates like glucose. They discovered that key metabolic pathways came together at a critical point to make the amino acid alanine, which effectively drives bacterial growth.

 

The results of the study identified new targets for small-molecule drugs to prevent C. difficile colonization and infection in the gut and offered a fresh method to quickly define microbial metabolism for other applications, such as the production of antibiotics and therapeutically and economically significant metabolites.

 

Researchers concluded that C. difficile significantly contributes to antibiotic-associated diarrhea and is the most common source of hospital-acquired infections. Understanding its cellular metabolic processes may help prevent and treat infections.
